Samenvatting Een veel voorkomende post-translationele modificatie die we in eiwitten terugvinden is de vorming van zwavelbruggen tussen cysteines. Wij vinden die zwavelbruggen vooral in extracellulaire eiwitten waar ze voor extra stabiliteit zorgen. Nu de aanmaak van recombinante eiwitten met verscheidene zwavelbruggen is niet altijd eenvoudig. Het proces waarbij zowel de juiste zwavelbruggen worden gevormd en het eiwit netjes wordt opgevouwen, noemen we oxidatieve eiwitvouwing. De meest populaire gastheer voor de productie van recombinante eiwitten is E. coli, maar eiwitten met meerdere zwavelbruggen zijn hier dikwijls misvouwen, gedegradeerd, of ze worden teruggevonden in ‘inclusion bodies’. Als gevolg hiervan hebben de alternatieve celvrije eiwitsynthese systemen op basis van een tarwekiemextract dan ook een opmars gekend en hebben het tot één van de standaard eiwitproductiemethoden gemaakt. Dit open cel-vrij-expressiesysteem maakt gebruik van de embryo’s van de tarwekiemen en is ideaal voor de expressie van eiwitten rijk aan zwavelbruggen, in het bijzonder voor eukaryote eiwitten. Doordat dit systeem van eukaryote oorsprong is, is het ook beter geschikt voor de productie eiwitten waarvan meerdere domeinen correct dienen te worden gevouwen. Verder heeft het systeem het bijkomend voordeel dat het een open systeem is waaraan een geoptimaliseerd mengsel van thiol-disulfide oxidoreductases en chaperones kan worden toegevoegd, zodat het eiwit geen kans maakt om te aggregeren of neer te slaan op het moment dat de polypeptideketen van de ribosomen rolt. Ik heb in mijn thesis een in vitro tarwekiemtranslatiesysteem geoptimaliseerd voor de expressie van drie belangrijke eukaryote eiwitten rijk aan disulfidebruggen. In het 1ste hoofdstuk geef ik een overzicht van de celvrije expressiesystemen met extra aandacht voor het E. coli en het tarwekiem expressiesysteem. De mechanismen van zwavelbrugvorming tijdens oxidatieve vouwingsproces en de betrokken chaperones en oxidoreductases worden besproken. De bestudeerde zwavelbrugrijke eiwitten worden voorgesteld en het doel van mijn thesis beschreven. In het 2de hoofdstuk toon ik de expressie van oplosbaar mFIZZ1. De expressie in het tarwekiemsysteem geeft oplosbaar, maar inactief mFIZZ1. Door humaan Quiescin

endoplasmatisch reticulum, aan dit open system toe te voegen kon ik monomerisch en biologisch actief eiwit bekomen met al zijn intra-moleculaire zwavelbruggen gevormd. Dit recombinante mFIZZ1 eiwit onderdrukt de productie van IL-5 en IL-13 in muis splenocyten gegroeid onder Th2 verdedigingscondities. Het hQSOX1b eiwit blijkt hier te functioneren als chaperon en als oxidase tijdens de oxidatieve eiwitvouwing. In het 3de hoofdstuk beschrijf ik hoe ik het in vitro tarwekiemtranslatie systeem optimaliseerde voor de expressie van het extreem toxische Androctonus australis hector (AahII) schorpioentoxine dat 4 disulfiden moet vormen. Ik toonde voor de eerste maal dat recombinant rAahII met een N-terminale GST-fusie in de oplosbare fractie terecht kwam. Tijdens het oxidatieve vouwingsproces moet dit toxine zowel disulfiden tussen cysteines in consecutieve als niet-consecutieve positie inbouwen, maar toch bleek dit proces onafhankelijk van de aanwezigheid van de oxidoreductases hQSOX1b en/of hPDI. injectie
in
muizenhersenventrikels. Het nanobodyTM NbAahII10 dat werd aangemaakt tegen het schorpioeneigen toxine herkende ook een structurele epitoop op het recombinante toxine, wat erop wijst dat het in vitro translatiesysteem correct gevouwen toxine produceerde. Daarenboven was dit nanobodyTM ook instaat om het recombinante toxine te neutraliseren bij alle geïnjecteerde muizen. Zo toonde ik voor de eerste maal het gebruik van een in vitro translatiesysteem voor de recombinante expressie van één van de meest toxische natriumkanaal-blokkerende schorpioentoxines in zijn biologisch actieve vorm. In het 4de hoofdstuk gebruikte ik het tarwekiemtranslatiesysteem om de invloed van oxidoreductases en chaperones op de expressie van de muizengroeifactor van het vasculaire endotheel (mVEGF164) te testen. mVEGF164 heeft niet minder dan 8 zwavelbruggen te vormen. Wat niet triviaal was voor de gastheer E. coli, lukte wel met het in vitro systeem, waar ik mVEGF164
met een N-terminale his-tag
gedeeltelijk in de oplosbare fractie tot expressie bracht. Acht inter- en intramoleculaire zwavelbruggen dienden correct te worden gevormd. Ook hier bracht coexpressie met verschillende concentraties aan hQSOX1b en/of hPDI geen soelaas. Met de optimale concentratie aan ‘Early Response to Dehydration’ chaperon
(ERD14) kon ik de relatieve hoeveelheid oplosbaar mVEGF164 verhogen. Het gezuiverde eiwit was biologisch actief en verhoogde het aantal gevormde bloedvaten in het membraan van een pas bevrucht kippenei, wat erop wijst dat het eiwit correct gevouwen is met al zijn disulfiden correct gevormd. In het 5de hoofdstuk geef ik een algemene conclusie en in het 6de en 7de hoofdstuk geef ik de samenvatting. De resultaten van mijn thesis zien er veelbelovend uit en ik hoop dat ze andere onderzoekers zullen aanmoedigen om naar het ideale chaperon en/of oxidoreductase mengsels te gaan zoeken voor de celvrije expressie van andere toxische eiwitten of zwavelbrugrijke eiwitten, in het bijzonder voor deze eiwitten waarvoor de klassieke expressiesystemen niet voldoen.
6 Summary Although disulfide bond formation in proteins is one of the most common types of post-translational modifications, the production of recombinant disulfide rich proteins remains a challenge. Most extracellular proteins are stabilized by multiple disulfide bonds formed by the oxidation of pairs of cysteine residues. Protein folding and disulfide bond formation are coupled, also known as oxidative protein folding. The most popular host for recombinant protein production is Escherichia coli, but disulfide rich proteins are here often misfolded, degraded, or found in inclusion bodies. As a consequence, the alternative cell-free protein synthesis based on wheat germ extract has become one of the standard protein production methods. This open system of cell-free expression using wheat germ embryos is ideal for the expression of disulfide rich proteins especially those from eukaryotes. This cell-free expression system is eukaryotic from origin, and as such, expected to synthesize multidomain proteins in a folded state. Also, it has the additional advantage that it is an open system in which the optimized mix of thiol-disulfide oxidoreductases or chaperones can be added, so that disulfide-rich proteins have no chance to aggregate the moment the polypeptide chain emerges from the ribosomes. In my thesis, I have optimized an in vitro wheat germ translation system for the expression of three important eukaryotic proteins that have to form multiple disulfide bonds.
In chapter 1, I introduce you to all the available cell-free expression systems in full details with a special focus on the wheat germ and E. coli extracts. The mechanism of disulfide bond formation during oxidative protein folding as well as the oxidoreductases and the chaperones involved are being discussed. Further, I present the disulfide rich proteins that I have been studying and the objectives of this study. In chapter 2, I show the soluble expression of Mouse Found in Inflammatory Zone (mFIZZ1). mFIZZ1 expression in wheat germ extract results in soluble, but biological inactive protein. However, expression in combination with human quiescin sulfhydryl oxidase (hQSOX1b), the disulfide bond-forming enzyme of the endoplasmic reticulum, results in soluble, intramolecular disulfide bonded, monomeric, and biological active protein. The recombinant mFIZZ1 protein clearly suppresses the production of the cytokines IL-5 and IL-13 in mouse splenocytes cultured under Th2 permissive conditions. The quiescin sulfhydryl oxidase hQSOX1b seems to function as a chaperone and oxidase during the oxidative folding. In chapter 3, I optimized an in vitro wheat germ translation system for the expression of the highly toxic Androctonus australis hector (AahII) scorpion toxin, which has to form four disulfide bonds. I showed for the first time that recombinant N-terminal GST-tagged rAahII toxin is solubly expressed in an in vitro translation system. Although four disulfide bonds between consecutive and non-consecutive cysteines need be correctly formed, co-translation with oxidoreductases (hQSOX1b or hPDI) did not increase the final yield. After proteolytical cleavage of the GST-tag, recombinant rAahII toxin showed a high toxicity with an LD50-value of 30 ng for mice after intracerebroventricular (icv) injection. Further, the nanobody NbAahII10 raised against the native AahII toxin recognizes a structural epitope on recombinant toxin, which indicates that rAahII expressed with a wheat germ in vitro translation system is correctly folded. Moreover, NbAahII10 neutralizes the toxicity of recombinant rAahII for all injected mice. I showed for the first time the recombinant expression of one of the most toxic sodium channel scorpion toxins in its biological active form using an in vitro translation system. In chapter 4, I used an in vitro wheat germ translation system as a tool to screen the function of oxidoreductases and chaperones on the expression of the mouse Vascular Endothelial Growth Factor (mVEGF164) protein, which has to form eight disulfide bonds. What was not possible for E. coli, seems to work for the in vitro translation
system. I showed that recombinant N-terminal His-tagged mVEGF164 is only partially solubly expressed. Screening with different concentrations of hQSOX1b and hPDI did not help to increase the solubility of mVEGF164. On the other hand, screening the expression with different concentrations of ERD14 helped to determine the optimal concentration to produce completely soluble mVEGF164. The produced mVEGF164 showed blood vessel formation activity on the membrane of a newly fertilized chicken egg, which suggests that all disulfide bonds are correctly formed. In chapter 5, I present the general conclusion of my work with the perspectives to follow-up this research study. Chapters 6 and 7 summarize my work. The results obtained during my doctoral thesis work should encourage the design of an appropriate chaperone and thiol/disulfide oxidoreductase-tuned wheat germ cell-free expression system for other challenging disulfide rich or toxic proteins, for which the more classical expression systems always fail.
