The PCR products are fragmented and labelled separately with two different reagents (Cy-3 and Cy-5 respectively) to allow the distinction between RHD and RHCE
An external control has been included to verify that the hybridisation has been performed correctly. This control will also help to place correctly the spot quantification grid. The chip also includes background hybridisation controls, which consist of spots containing solvent alone (50% DMSO) to allow the determination of background hybridisation.
Labelled PCR p products will be applied pp to the microarray surface for hybridisation. Confocal scanner detects the bound DNA as fluorescence is emitted by the fluorophores upon excitation with a laser beam. 300

Conclusions (1) • Three genotypes described here were not found in „Rhesus Base“ and not reported in the literature • Case 2 serology gy correspond p to weak D • Cases 1 and 3 have normal D qualitative and quantitative serology results • Amino acids exchanges in Case 1 and 2 correspond to RhCcEe protein • cDNA sequencing of Case 3 should be done to clarify exon 9 sequence
Conclusions (2) • Case 1 could be hybrid gene RHCE(1)-RHD(2-10) • Case 2 could be either point mutation or hybrid gene with smaller RHCE exchange g in exon 5 • Case 3 could be hybrid gene RHD(1 RHD(1-8)-CE(9)-D(10) 8) CE(9) D(10) or
RHD(1-7)-CE(8-9)-D(10) … hypothesis
• RhC specific aa Cysteine in position 16 does not influence RhD expresion • RHD specific sequence in exon 9 is not necessary for normal RhD expression
