RIWAYAT HIDUP Penulis dilahirkan di Jakarta pada tanggal 22 Mei 1984 sebagai anak kedua dari dua bersaudara, putra dari pasangan Gunawan dan Lily. Penulis lulus pada tahun 2002 dari SMU Negeri 26 Jakarta dan diterima di Institut Pertanian Bogor (IPB) melalui jalur Seleksi Penerimaan mahasiswa Baru (SPMB). Penulis memilih Program Studi Biologi, Departemen Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam. Selama mengikuti perkuliahan, penulis juga pernah aktif sebagai anggota PAMABI (Paguyub an Mahasiswa Biologi).

ABSTRAK R.M.RAGA BIMO S. Uji Keseragaman Famili F8 Kedelai Dari Persilangan Kultivar Slamet dengan Wase. Dibimbing oleh SUHARSONO dan MUHAMMAD JUSUF. Tujuan penelitian ini adalah menguji keseragaman genetik berdasarkan produksi biji dari famili-famili kedelai generasi F8 dari persilangan kultivar Slamet dengan Wase. Famili yang diuji terdiri dari 24 famili. Kultivar Slamet digunakan sebagai pembanding. Percobaan dilakukan dengan menanam kedelai dengan jarak tanam 40 x 20 cm, dengan 2 biji tiap lubang. Setiap famili terdiri dari 80 biji, dan disisipi dengan kultivar Slamet. Analisis ragam produksi biji pada generasi F8 dari persilangan Slamet dengan Wase menunjukkan bahwa 22 famili telah seragam dan 2 famili belum seragam secara genetik. Dari 22 famili yang seragam terdapat satu famili yang memiliki produksi lebih tinggi daripada Slamet yakni FSW22. Galur ini dapat dijadikan sebagai galur harapan.
ABSTRACT R.M. RAGA BIMO S. Test of Homogenity F8 family of soybean from crossing between Slamet and Wase Cultivars. Under direction of SUHARSONO and MUHAMMAD JUSUF. The aim of this experiment is to test the genetic homogenity and the productivity of the F8 generation families of crossing between Slamet and Wase Cultivars. The Families tested consisted of 24 families. Slamet cultivar was used as a standard of comparison. The experiment was conducted by planting soybean seeds in the planting distance of 40 cm x 20 cm, 2 seeds per hole. From each family 80 seeds were planted, and Slamet cultivar was inserted in between five families. Variance an alysis of seed production of F8 family from crossing between Slamet and Wase showed that 22 families were homogen and 2 families were not genetically homogen yet. From 22 homogen families, one family, i.e. FSW 22, had higher productivity than Slamet. This family can be used as potential lines.

PENDAHULUAN Kedelai merupakan sumber protein nabati yang sangat penting baik untuk pangan maupun pakan dan mempunyai nilai ekonomi yang tinggi bagi Indonesia. Kedelai termasuk tanaman semusim yang berbatang perdu dengan tinggi antara 30-100 cm, dapat bercabang sedikit atau banyak. Buah kedelai berbentuk polong yang berisi 1-5 biji, tetapi sebagian besar berisi 2-3 biji (Sumarno 1984). Biji kedelai mengandung 35% protein, 18% lemak, 35% karbohidrat, dan 8% air (Maesen 1992). Di Indonesia, kedelai digunakan sebagai bahan baku industri pembuatan tempe, tahu, tauco, kecap, susu kedelai, dan bahan makanan ternak. Tingkat konsumsi nasional kedelai terus naik seiring dengan meningkatnya jumlah penduduk. Produksi kedelai Indonesia pada tahun 2003 baru mencapai 671 600 ton (BPS 2004), sementara kebutuhannya mencapai 2 juta ton per tahun (Kompas 2004). Kekurangan tersebut harus dipenuhi dengan impor yang dapat mencapai 1.2 juta ton per tahun (Kompas 2004). Oleh karena itu, diperlukan berbagai usaha untuk meningkatkan produksi kedelai. Salah satunya adalah peningkatan produksi dengan menggunakan varietas unggul. Varietas unggul dapat diperoleh dengan cara persilangan antar varietas yang telah ada dan sudah teruji baik serta mampu beradaptasi dengan kondisi lapang (Boer 1989). Usaha peningkatan yang lain adalah dengan ekstensifikasi atau perluasan lahan pertanian kedelai di luar pulau Jawa. Tetapi, usaha tersebut menghadapi kendala dengan kondisi lahan marginal. Salah satu lahan marginal di Indonesia ialah lahan asam yang mengandung kadar alumunium yang tinggi (Notohadiprawiro 1983). Keracunan alumunium (Al) akan menghambat pertumbuhan akar primer dan menghalangi pembentukan akar lateral dan bulu akar. Keracunan Al menyebabkan ujung akar menebal, berwarna coklat seperti busuk dan mengering sehingga menghasilkan sistem perakaran tanaman yang kerdil dan pendek, karena terjadi penekanan terhadap perkembangan jaringan meristem akar (Sanchez 1976 dalam Rustianti R 1994). Selain itu alumunium dapat menghambat pertumbuhan bintil akar untuk mengikat Nitrogen (Sunarto 1989). Kenaikan hasil merupakan tujuan utama dari pemuliaan tanaman. Hal ini dapat dilakukan dengan menyediakan kultivar yang
lebih produktif (Allard 1995). Seleksi merupakan inti pekerjaan dari pemuliaan tanaman. Seleksi adalah proses pemilihan genotipe-genotipe unggul dari suatu populasi untuk membentuk populasi generasi selanjutnya yang lebih baik. Keberhasilan perbaikan populasi ini sangat bergantung pada keberhasilan memilih genotipe-genotipe yang baik. Seleksi dilakukan untuk meningkatkan nilai tengah populasi dan menurunkan ragamnya. Untuk mendapatkan kultivar unggul, beberapa persilangan antara kultivar Slamet dan kultivar lainnya telah dilakukan. Salah satunya adalah persilangan antara kultivar Slamet dan Wase (Paserang 2003). Dari persilangan ini diharapkan mendapatkan keturunan yang berproduksi tinggi, berbiji besar dan toleran terhadap tanah asam. Kultivar Slamet merupakan kultivar unggul yang memiliki produksi tinggi (2.26 ton/ha), berbiji yang berukuran sedang (12.5 g/100 biji), dan toleran terhadap asam (Sunarto 1995). Deskripsi lengkap kultivar Slamet disajikan pada lampiran 1. Sedangkan Wase merupakan kultivar dari Jepang yang mempunyai biji berukuran besar (20.56 g/100 biji). Keturunan dari persilangan ini telah dilakukan seleksi sampai dengan F7 (Suharsono et al. 2003, Abdillah 2004, Supriyadi 2005, Fathony 2006). Uji keseragaman genetik terhadap generasi F7 menunjukkan bahwa belum semua famili adalah seragam secara genetik (Fathony 2006) sehingga perlu diadakan seleksi lebih lanjut. Pada penelitian ini uji keseragaman genetik dilakukan pada lahan yang tidak dimaksudkan untuk seleksi terhadap tanaman yang toleran lahan asam. Uji keseragaman genetik pada penelitian ini didasarkan pada produksi biji. Tujuan Penelitian ini bertujuan untuk menguji keseragaman genetik famili F8 dari persilangan kultivar Slamet x Wase. Waktu dan Tempat Penanaman dilaksanakan mulai bulan Desember 2005 sampai dengan Maret 2006 di Kebun Percobaan IPB Pagentongan dan pengolahan pascapanen dilakukan di Laboratorium Biologi Seluler dan Molekuler Tanaman, Pusat Penelitian Sumberdaya Hayati dan Bioteknologi, Kampus IPB Darmaga.

BAHAN DAN METODE Bahan dan Alat Tanaman yang digunakan adalah 24 famili terpilih generasi F8 dari persilangan Slamet X Wase (Fathony 2006) dan kultivar Slamet digunakan sebagai pembanding keseragaman genetik. Metode Penanaman Sebanyak 24 famili F8 terpilih dan 1 kultivar pembanding ditanam di lapang. Sebanyak 80 biji dari tiap famili ditanam 2 biji per lubang, dengan jarak tanam 40 cm antar baris dan 20 cm dalam baris tanaman. Famili diselingi dengan kultivar Slamet sebagai pembanding. Denah penanaman disajikan pada lampiran 2. Pemeliharaan Pemupukan dasar yang diberikan ialah 150 kg/ha Urea, 100 kg/ha TSP dan 100 kg /ha KCl dengan cara ditanam di antara barisan tanaman. Pemberian urea diberikan dua tahap yaitu pada saat tanam dan penyiangan pertama. Penyiangan dilakukan dua kali yaitu pada 3 MST (minggu setelah tanam) dan 7 MST. Pengendalian hama dilakukan dengan pemberian Furadan ketika penanaman dan penyemprotan dengan Akodan tiap minggu dari 2 MST s/d 8 MST. Pemanenan Pemanenan dilakukan ketika 90% polong sudah berubah warna menjadi kuning kecokelatan. Cara pemanenan dengan mencabut seluruh tanaman kemudian dimasukkan ke dalam kantung kertas. Setiap kantung berisi satu tanaman. Setelah itu tanaman dijemur hingga beberapa polongnya pecah. Untuk mengupas polong menjadi biji, tanaman dipukul - pukul dengan kayu hingga bijinya lepas dari polong. Lalu, biji - biji yang diperoleh ditampi dan dibersihkan dari kotoran. Pengamatan Karakter - karakter yang diamati pada setiap famili adalah umur mulai berbunga, umur panen (hari), t inggi tanaman (cm), jumlah buku subur, jumlah cabang, bobot 100 biji (g), jumlah polong, produksi biji tiap tanaman, warna bunga. Analisa Data Data produksi biji dianalisis dengan menggunakan perangkat lunak SPSS (Statistical Product Service Solution). Analisis data yang dilakukan meliputi analisis nilai tengah setiap famili, koefisien
keragaman dan uji keseragaman setiap famili dengan rumus : n

HASIL DAN PEMBAHASAN Uji Keseragaman Uji keseragaman genetik dapat dilihat dengan membandingkan ragam antara famili dari hasil persilangan dan tet uanya, yaitu kultivar Slamet. Kultivar Slamet digunakan sebagai pembanding keseragaman genetik karena sudah dilepas sebagai kultivar nasional dan setiap kultivar yang sudah dilepas adalah seragam secara genetik. Uji keseragaman famili F8 dengan Uji F dari persilangan Slamet x Wase berdasarkan produksi biji tiap
tanaman menunjukkan bahwa belum semua famili F8 ser agam. Dari 24 famili yang terpilih (F8) hanya 22 famili yang telah seragam secara genetik. Dengan demikian secara genetik 22 galur ini tidak akan bersegregasi lagi. Keseragaman ini ditunjukkan oleh masing-masing ragam dari 22 famili yang tidak berbeda nyata dari ragam Slamet pada taraf uji 5%, sedangkan 2 famili lainnya masing-masing mempunyai ragam yang berbeda nyata dengan ragam populasi kultivar Slamet pada taraf uji 5 % (Tabel 1)

Secara teori, keragaman genetik pada generasi F8 sangatlah kecil. Poespodarsono (1988), mengungkapkan bahwa p ada tanaman menyerbuk sendiri termasuk kedelai, akan terjadi penurunan frekuensi heterozigot menjadi setengah pada setiap generasi, sehingga pada generasi F8 akan memiliki frekuensi heterozigot (1/2)6. Pada percobaan ini, masih beragamny a 2 famili tersebut diatas pada generasi F8, diduga disebabkan oleh adanya pengaruh lingkungan, yakni kesuburan tanah. Keragaman yang muncul akibat pengaruh lingkungan ini dapat dilihat dari besarnya koefisien keragaman kultivar Slamet. Koefisien keragaman dapat digunakan untuk membandingkan keragaman dua atau lebih populasi, meskipun satuan pengukurannya tidak sama (Walpole 1990). Ragam kultivar Slamet adalah 34.56 dengan nilai koefisien keragaman yang tinggi yakni 64.29% (Tabel 1). Kultivar Slamet seharusnya memiliki nilai koefisien keragaman dibawah 20%, yakni nilai batas yang dapat diterima dalam pelepasan kultivar kacang-kacangan (Suhartina 2003). Besarnya nilai koefisien keragaman melebihi 20% bukan disebabkan pengaruh genetik melainkan oleh faktor keragaman lingkungan, yang kemungkinan hal ini disebabkan oleh keragaman kesuburan tanah. Berdasarkan observasi dilapang, lahan yang digunakan tersebut memiliki permukaan tanah yang miring. Hal ini mengakibatkan terjadinya erosi pada tanah yang disebabkan oleh aliran air hujan. Curah hujan yang cukup tinggi menyebabkan pemupukan yang dilakukan tidak efektif karena pupuk akan terbawa air hujan kemudian menumpuk pada tanah yang permukaannya lebih rendah dan nutrisi – nutrisi yang dibutuhkan oleh tanaman dapat tercuci. Akibatnya pada tanah yang pemukaannya lebih rendah kondisinya lebih subur dibandingkan dengan tanah yang permukaannya lebih tinggi. Uji Nilai Tengah Produksi Pengujian nilai tengah produksi terhadap 24 famili generasi F8 menunjukkan bahwa sebanyak satu famili memiliki nilai tengah yang lebih tinggi daripada kultivar Slamet yaitu FSW 22 (Tabel 1). Berdasarkan uji keseragaman genetik, FSW 22 adalah seragam secara genetik. Oleh sebab itu famili FSW 22 dapat dijadikan sebagai galur harapan karena sudah seragam dan berproduksi lebih tinggi daripada Slamet.
Sebelum dilepas sebagai kultivar nasional galur ini diuji daya hasilnya pada berbagai lokasi. Selain uji keseragaman dan produksi biji, seleksi juga dilakukan terhadap ukuran biji yakni bobot per 100 biji (g/100 biji). Biji kedelai digolongkan dalam tiga kategori yaitu kecil (6-10 g/100biji), sedang (11-12 g/100biji), dan besar (13 g atau lebih/100 biji) (Sumarno 1984). Dengan dasar ini enam famili F8 berbiji sedang dan delapanbelas famili berbiji kecil serta tidak ada yang berbiji besar (Tabel 1). Pada percobaan ini kultivar Slamet mempunyai biji berukuran 8.04 g/100 biji yang lebih kecil dari potensinya yaitu 12.5 g/100 biji (Suhartina 2003). Jika dibandingkan dengan percobaan Supriyadi (2005), ukuran biji kultivar Slamet pada penelitian ini juga lebih kecil. Kecilnya ukuran biji kemungkinan disebabkan oleh tidak optimumnya kondisi pertumbuhan yang meliputi curah hujan, iklim, keasaman tanah, dan kandungan unsur hara tanah. Bila kondisi pertumbuhan optimal, ukuran biji kultivar Slamet akan sama atau mendekati potensinya. Demikian juga dengan famili yang diuji, ukuran bijinya akan lebih tinggi dari pada hasil percobaan ini. Curah hujan selama pertumbuhan tanaman pada percobaan ini yaitu antara Januari dan Maret 2006 cukup tinggi yakni sekitar 252-640 mm/bulan dengan 24-46 hari hujan tiap bulan (Lampiran 3). Untuk mendapatkan produksi tinggi, kedelai membutuhkan curah hujan 150 – 200 mm/bulan selama masa pertumbuhan dan kurang dari 50 mm/bulan selama masa pematangan. Pada musim kemarau dengan pengairan yang cukup kedelai dapat menghasilkan rata–rata 1.97 ton/ha sedangkan pada lokasi yang sama pada musim hujan hanya menghasilkan 0.61 ton/ha (Baharsjah et. al 1985 .. Curah hujan yang tinggi biasanya diikuti dengan men urunnya lama penyinaran. Lama penyinaran khususnya pada saat pengisian dan pemasakan biji sangat berpengaruh terhadap produksi kedelai. Pada percobaan ini, curah hujan pada saat pengisian dan pemasakan biji yaitu pada bulan Februari dan Maret 2006 sangat tinggi. Curah hujan yang tinggi juga diyakini menyebabkan reaksi tanah menjadi masam akibat kehilangan basa-basa karena mudah tercuci (Nyakpa et. al 1988). Berdasarkan analisis tanah yang dilakukan menunjukkan bahwa tanah pada area penanaman termasuk tanah masam dengan pH berkisar 5.0 (Lampiran 4). Tanah yang masam

mengandung kadar alumunium yang tinggi sehingga menyebabkan kemampuan tanah untuk mengikat unsur-unsur hara menjadi berkurang. pH tanah yang paling baik untuk pertumbuhan kedelai adalah 6.8. Namun pH tanah 5.5-6.0 sudah dianggap cukup. Rendahnya pH tanah dapat mempengaruhi pertumbuhan kedelai (Ismail & Suryatna 1984). Berdasarkan uji keseragaman genetik, produksi biji tiap tanaman dan ukuran biji bila dibandingkan dengan kultivar Slamet, keduapuluhempat famili F8 dapat dikelompokkan menjadi enam kelompok (Tabel 2). Satu famili memiliki produksi biji lebih tinggi daripada Slamet dan telah seragam secara genetik tetapi berukuran biji kecil yaitu FSW 22, sehingga FSW 22 dapat dijadikan galur harapan. Satu famili berproduksi sama dengan Slamet, berbiji sedang, dan seragam yaitu FSW 21. Tujuh famili berproduksi sama dengan Slamet, seragam tetapi berbiji kecil. Lima famili berbiji sedang, seragam tetapi berproduksi lebih kecil daripada Slamet. Delapan famili berproduksi lebih kecil, berbiji kecil, dan seragam. Dua famili memiliki produksi biji sama dengan Slamet dan belum seragam secara genetik serta berbiji kecil. Karakteristik setiap individu tanaman dari setiap famili disajikan pada lampiran 5.
SARAN Uji kemantapan produksi perlu dilakukan terhadap FSW 22 pada berbagai lokasi.
