Produksi Sel Somatis Sebagai Donor Transfer Nukleus Untuk Menghasilkan Embrio Kloning Kambing Gatot Ciptadi1), Trinil Susilawatil) Arief Boediono 2), Moch. Sasmiti Djati3), Sutiman B. Sumitro 3) 1) Fakultas Peternakan, Universitas Brawijaya, Malang. 2) Fakultas Kedokteran Hewan IPB Bogor 3)Fakultas MIPA, Universitas Brawijaya, Malang Abstrak Diterima tanggal 5 September 2005 Disetujui tanggal 18 Mei 2006 Penelitian dilakukan untuk mempelajari sistem preparasi dan produksi beberapa jenis sel somatis untuk sel donor dalam produksi embryo kloning dengan transfer inti menggunakan metode injeksi intra sitoplasma (TNIS). Diproduksi tiga jenis sel donor yaitu sel fibroblast fetus, sel kumulus dan fibroblas kulit telinga kambing dewasa. Semua jenis sel donor setelah dilakukan 2-3 passase ditransfer ke dalam sel resipien oosit hasil maturasi in vitro. Donor sel diseleksi berdasarkan ukuran sel, yaitu ukuran kecil (G0/G1) yang digunakan dalam transfer nukleus. Aktivasi kimia dilakukan dengan menggunakan Ca ionophore A-23187 selama 7 menit dan dilanjutkan dengan 6-DMAP selama 4 jam. Fetal fibroblast sementara ini merupakan jenis sel yang terbaik, diikuti oleh sel kumulus dan fibroblast kulit telinga kambing dewasa, karena mempunyai daya reproductivitas dan kemampuan reprograming yang paling tinggi sebagai donor inti. Production Of Somatic Cells As A Donor Cels For Nuclear Transfer On Reconstructed Of Cloned Goat Abstract Production of somatic cells as a donor eels for nuclear transfer on reconstructed of cloned goat This study was done to produce a goat nuclear transfer embryos by optimizing a several donor cell preparation and production. Fetal fibroblast, cumulus cells and skin ear adult fibroblast were used as donor cells. A comparaison of efficacy among donor somatic cells of differents types of donor cells, were transferred to enucleated oocytes matured in vitro after 2-3 time subculturing. Donor cells selected was small size (G0/G1). Activation of reconstructed oocyte was conducted using Ca Ionophore A-23187 for 5 minute followed by 6-DMAP for 4 hours. Fetal fibroblast were the most effective among examined cell because its good reproducibility and reprogramming as donor karyoplast. Keywords: donor cells, nuclear transfer, activation, goat
