Perlunya pencegahan Pencegahan kanker sangat penting bagi masyarakat dan perekonomian negara. Pengobatan kanker jauh lebih mahal dibandingkan pencegahannya. Kanker bisa dicegah dengan menghindari rokok, mengurangi polusi, radiasi, infeksi, dan mengkonsumsi makanan bergizi dalam jumlah yang cukup, konsumsi sayuran dan buahan /hari, kakao

Metode in vitro untuk menguji aktivitas antikanker pangan dapat dilakukan dengan menggunakan sel-sel kanker yang dikultur dengan ekstrak dari bahan uji dalam media sintetik yang mengandung semua zat dibutuhkan oleh sel kanker untuk hidup secara optimal Sel-sel kanker dapat diperoleh langsung dari jaringan kanker, baik dari manusia maupun dari hewan. Sel yang diperoleh disebut secara langsung disebut sel kanker primer

• Sel-sel kanker dapat juga dibeli dalam bentuk alur sel (cell line), yaitu sel primer yang telah dipelihara secara in vitro dan telah mengalami perbanyakan, setiap alur sel diberi nama masing-masing sesuai dengan identitas sumbernya,.misalnya: K562 adalah sel kanker yang berasal dari kanker darah manusia. Sel Hela adalah selyang berasal dari jaringan kanker jaringan mulut rahimmanusia. • Contoh penelitian evaluasi aktivitas antikanker pangan dalap diklik pada evaluasi antikanker1 Pada contoh penelitian ini dapat diikuti metode kultur sel kanker secara bertahap dan interpretasi hasilnya. Sebagai pembanding digunakan juga sel limfosit manusia untuk membandingkan aktivitas zat uji terhadap sel normal nonkanker. Zat anti kanker yang ideal adalah yang mampu membunuh atau menghambat pertumbuhan sel-sel kanker tetapi tidak menghambat, membunuh atau mengganggu pertumbuhan sel normal dalam tubuh.
Peningkatan sistem imun: immunostimulan, immunoenhancer, immunomodulator Sistim imun: • sistim kekebalan tubuh • sel-sel imun (sel darah putih: limfosit B/T, monosit/makrofag, eosinofil/basofil/mastosit • senyawa yang dihasilkan oleh sel-sel imun: antibodi, inteferon, sitokin Evaluasi aktivitas immunomodulator bahan pangan secara in vitro dapat dilakukan melalui kultur sel-sel imun.

Sel limfosit dapat berproliferasi: kultur sel limfosit untuk menentukan kemampuan sel untuk berproliferasi. Makin mampu sel berproliferasi makin baik aktivitas zat uji. Stimulasi fungsi makrofag Makrofag adalah salah satu anggota sel darah putih yang berfungsi sebagai sel penelan (fagosit) mikroorganisme (mo) (bakteri,virus) yang tidak diinginkan yang masuk ke dalam tubuh. Setelah menelan, sel makrofag harus dapat membunuh mo yang telah ditelannya. Mekanisme makrofag membunuh mo yang ditelannya adalah melalui semburan radikal bebas dan hidrolisis oleh enzim-enzim. Makin baik fungsi makrofag makin sulit bagi mo untuk hidup dalam tubuh kita. Makanan yang tepat dapat memperbaiki fungsi makrofag.

Evaluasi aktivitas hormon secara in vitro Hormones: • substances secreted into blood vessels by certain producing glands (endocrine system) : dalam jumlah sangat sedikit tapi efek nyata i.e. female sex hormone ( its producing gland is clear (ovary) and the target gland is also clear (female reproductive system). Produces effect with small quantity which is sexual development. • Organs: the liver, kidney, adrenal gland, central nervous system, immune system, cardiovascular system, and bones are target tissues of the steroid hormones. Phytoestrogens (estrogen tanaman) terdapat pd tanaman. Meliputi: isoflavones, lignins, coumestans, or resorcylic acid lactones that are structurally similar to the mammalian estrogen and have a weak estrogenic potency ranging from 1/500 to 1/1000 that of 17ß-estradiol. Hormon dr makanan (exohormones) dpt berinteraksi dgn hormon dr dlm tubuh (endogenous): 1. influence hormone synthesis—the release, the transport, the effect, the metabolism, and the excretion of hormones. 2. interferes with the hormones at receptors. Phytoestrogens such as coumestrol, daidzein, and genistein; medications such as diethylstilbestrol, ethinyl estradiol, and tamoxifen; and industrial chemicals such as DDT, p-nonylphenol, and bisphenol A belong to this group. They bind to the estrogen receptor and interact with the binding of the hormone. 3. Block receptors, ie, the receptors for the male hormone testosterone. DDT metabolite p,p`-DDE or vinclozoline metabolites: antagonists. Substances that compete with the physiologic ligand, eg, a hormone at the receptor, and imitate (meniru) its effect are called
agonists;

The in vitro tests are able to identify hormone-like potential
of a chemical and its relative potency. However, they do not include toxicokinetics and its metabolites, so results have to be verified by in vivo testing. 1.The recombinant yeast estrogen assay -uses yeast cells yg telah diberi gen penghasil reseptor estrogen manusia, (estrogen responsive elements and lac-Z reporter gene encoding ß-galactosidase). The cells are incubated dgn senyawa uji bersama dgn senyawa pewarna (chromogen). Senyawa uji yg dapat berikatan dgn reseptor dapat memicu enzim ß-galactosidase, yg merubah warna chromogen to yellow lalu intensitas warna diukur dengan spectrophotometer.28 2. E-screen test: the human MCF-7 breast cancer cell line is incubated with the test compound for several days to allow proliferation. Thereafter, the number of cells is quantified, eg, by measuring optical density.

3. estrogen-R() competitor screening test: the human estrogen receptor is fixed to 96 wells. The test compound and fluorescein-labeled 17ß-estradiol are added. After incubation and washing, the fluorescence of 17ß-estradiol (atau senyawa uji) attached to the receptor is measured.30 4.Placental aromatase assay: uses aromatase cytochrome P450–mediated conversion of C19 androgens to aromatic C18 estrogens. Androgens have been shown to inhibit this reaction.31 5. The androgen binding or reporter gene assay and the thyroid receptor binding assay: Menentukan affinity relative dr senyawa uji (androgen-like or thyroid hormone–like) terhadap reseptor androgen dibandingkan dengan endogenous hormones.
