Replikasi Secara konsep p sederhana Proses mekanismenya Æ komplek Kesederhanaannya Æ krn konsep dr Watson & Crick Transfer a s e informasi o as melibatkan e bat a pembukaan pe bu aa double doub e helix DNA yang diikuti secara bersamaan dengan pembentukan dua pita baru pasangan dari pita DNA yang lama
Mekanisme pengkopian DNA Æ Mekanisme pengkopian DNA Æ melibatkan pembukaan double helix Setiap rantai menjadi pola / templat untuk pita baru p p
pp g DNA 1. “Unzipping” - Helicase unwinds the DNA (it makes it flat flat, so it’s it s not twisted anymore) - Hydrogen H d b bonds d b break k between the base pairs - “Replication Fork” = the place e e st strands a ds sta startt to sepa separate ate where
2 “Parent 2. Parent Strands Strands” act like a template (they act like a guide, so the matching bases know where to go) 3. DNA Polymerase (the “helper”) - puts ‘free’ bases on the parent strands - the bases form new strands of DNA 4. Finished! Now there are two identical pieces of DNA
* “polymer” polymer = a chain of many similar pieces (DNA is a polymer polymer. It is a chain of nucleotides nucleotides.)) `
DNA Polymerase –
- it creates a polymer of DNA - it proofreads the new DNA p `
DNA Helicase - it unwinds the double helix

Translation - animation
Not all codons are used with equal frequency. There is a considerable amount of variation in the patterns of codon usage between different organisms. i

Only tRNAfMet is accepted to form Twothe initiation initiation factors complex complex. (IF1 &IF3) bind to a 70S All ribosome. further charged tRNAs require Æpromote fully assembled the dissociation (i.e., 70S) of 70S ribosomes ribosomes into free 30S and 50S subunits. The Shine-Dalgarno sequence mRNA and ÆIF2, helpwhich ribosomes and carries mRNA aligns correctly for the - GTP start of translation. - the charged tRNA Ribosome consists of Æbind a free 30S subunit. -A site Ætoaminoacyl P site After Æthese peptid have l all -Æ peptidyl - bound, E site Æthe exit30S initiation complex is complete.
•First the mRNA attaches itself to a ribosome (to the small subunit). •Six bases of the mRNA are exposed exposed. •A complementary tRNA molecule with its attached amino acid (methionine) base pairs via its anticodon UAC with the AUG on the mRNA in the first position P. •Another tRNA base pairs with the other three mRNA bases in the ribosome at position A. •The enzyme peptidyl transferase forms a peptide bond between the two amino acids. •The first tRNA (without its amino acid) leaves the ribosome.
The ribosome moves along the mRNA to the next codon (three bases). The second tRNA molecule moves into position P. Another tRNA molecule pairs with the mRNA in position A bringing its amino acid. A growing polypeptide is formed in this way until a stop codon is reached.
Peptide P tid bond b d formation Æ catalyzed by an enzyme complex called peptidyltransferase Peptidyltransferase consists of some ribosomal proteins and the ribosomal RNA Æ acts as a ribozyme. The process is repeated until a termination signal is reached.
A stop codon on the mRNA is reached and this signals the ribosome to leave the mRNA. A newly synthesised protein is now complete!
Termination of t translation l ti occurs when one of the stop codons (UAA, UAG, or UGA) appears in the A site of the ribosome ribosome. No tRNAs correspond to those sequences sequences, so no tRNA is bound during termination. Proteins called release factors participate in termination
