Egy kutató munkájának jelentőségét kifejezheti az, hogy műveire összesen hány mások által írott hivatkozást kap. Az önhivatkozásoknak (vagyis mikor a szerző egyik művében saját másik művére utal) e szempontból általában nem tulajdonítanak jelentőséget. History 1 1907 W. A. Noyes became the first editor of the new publication, Chemical Abstracts, which appeared in January 1907. CA was first published at the U. S. Bureau of Standards. In its first year of publication CA contained a total of fewer than 12,000 abstracts. By contrast, CA published a million abstracts in 2006.

History 5 1965 A new era dawned with the introduction of the CAS Chemical Registry System in 1965. Using the CAS Registry Number® to identify substances without the ambiguity of chemical nomenclature, the CAS REGISTRY has proved to be a boon to chemical research, health and safety information, and the communication of chemical information in many media.

History 6 1966 CAS management and technical teams devised an automated processing system that not only produced printed CA issues and indexes more efficiently but also fed a computer-readable database that could generate new kinds of services. CAS was an early adopter of the new photocomposition technology, and went on to produce services in print, microform, and magnetic tape.

History 7 1968 CAS applied the available technology to capture chemical information in computer-readable form. For example, operators keyed substance information into the Mohawk 1181 data recorder for storage on magnetic tape, which was later fed into the mainframe computer.

History 8 1980 CAS ONLINE was launched in 1980, making it possible for users (primarily information specialists) to search the CAS Registry database. Users with a specific model of intelligent graphics terminal, the Hewlett-Packard 2647A, could select structure features from a menu and then assemble them on the terminal monitor using a graphics tablet and stylus. These terminals could display answers with well-drawn structure diagrams.

What is Chemical Abstract Registration number (CASRN)? CAS Registry Numbers (often referred to as CAS RNs or CAS Numbers) are unique identifiers for chemical substances. A CAS Registry Number itself has no inherent chemical significance but provides an unambiguous way to identify a chemical substance or molecular structure when there are many possible systematic, generic, proprietary, or trivial names. CAS RN 1261106-14-2 is the most recent CAS Registry Numbe CAS Registry Numbers are used in many other public and private databases as well as chemical inventory listings and, of course, are included in all CAS-produced databases. 31

Kémiai szerkezet kódolása ASCIIkarakterekkel: SMILES A SMILES betűszó, az angol simplified molecular input line entry specification (egyszerűsített, begépelhető molekula-leíró rendszer) rövidítéséből ered, Arthur és David Weininger fejlesztette ki az 1980-as években1,2. Széles körben használt leíró nyelv a molekulák ASCII-karakterekkel történő tömör kódolására. A legtöbb molekula-rajzoló és kémiai adatbázis program elfogadja bemenetként a SMILES kódokat, és képes azokat kétdimenziós rajzokká.

PDB identification code Every molecular model (atomic coordinate file) in the Protein Data Bank (PDB) has a unique accession or identification code. These codes are always 4 characters in length. The first character is a numeral, while the last three characters can be either numerals or letters. There are over 400,000 possible 4-character PDB identification codes (419,904 or 466,560 if "0" is allowed as the first character). Thus, the 51,000 entries in 2008 have used up less than 12% of the available codes. Isolated from the pacific yew tree
Paclitaxel is a mitotic inhibitor used in cancer chemotherapy. It was discovered in a National Cancer Institute program at the Research Triangle Institute in 1967 when Monroe E. Wall and Mansukh C. Wani isolated it from the bark of the Pacific yew tree, Taxus brevifolia and named it 'taxol'. When it was developed commercially by Bristol-Myers Squibb (BMS) the generic name was changed to 'paclitaxel' and the BMS compound is sold under the trademark 'Taxol'.

Sildenafil Sildenafil (compound UK-92,480) was synthesized by a group of pharmaceutical chemists working at Pfizer's Sandwich, Kent research facility in England. It was initially studied for use in hypertension (high blood pressure) and angina pectoris (a form of ischaemic cardiovascular disease). Phase I clinical trials under the direction of Ian Osterloh suggested that the drug had little effect on angina, but that it could induce marked penile erections.

Induced-Fit Model However, the lock-and key model did not take into account the conformational changes that occur for both the ligand and the target macromolecule. An extension of this model was proposed by Daniel Koshland in 1958 and called the „induced-fit theory”. This theory proposed that in the recognition process both ligand and target mutually adapt themselves by small conformational changes until an optimal fit is achieved.

High throughput screening (HTS) The attempt to replace the quality of scientific arguments by the sheer quantity of data as expressed in HTS or ultra-HTS in the past has failed. An approach that is based on a much broader understanding of biochemical and genetic mechanisms of diseases appears to represent the necessary correction.

Cost of HTS
It is ca. 3.17$/compound screened (labor and facility costs represent the greater proportion than consumable and reagents). A large pharma prosecutes ca. 65 HTS campaigns (400.000 compounds per screen) yearly; it costs 82.4$! (ca 10 % of the total cost of bringing a NCE to market) 1:67 survival rate for early stage research projects achieving new drug application (NDA) – Successes?! It is predicted that an annual yield of 0.3 NCE per company (23 large pharma companies) will result from current HTS activities. This level is an order of magnitude below the current predictions for sustained pharma profitability... Nevirapine (Boehringer) is the one cited example that was discovered as a result of HTS. ‘The reasons for deficit are cultural and organizational’ (Mark Beggs et al.)

LEAD compound One of the key-events in DD is finding a lead compound ‘Ein Königreich für eine neue Leitstruktur’ A lead compound (i.e. the "leading" compound, not lead metal) in drug discovery is a chemical compound that has pharmacological or biological activity and whose chemical structure is used as a starting point for chemical modifications in order to improve potency, selectivity, or pharmacokinetic parameters. Lead compounds are often found in high-throughput screenings ("hits") or are secondary metabolites from natural sources. Newly invented pharmacologically active moieties may have poor druglikeness and may require chemical modification to become drug-like enough to be tested biologically or clinically.

Lipinski's Rule of Five The ‘rule-of-five’ devised by Lipinski and coworkers at Pfizer from an analysis of 2245 drugs from the WDI believed to have entered Phase II trials. The rule-of-five generates an alert (indicating possible absorption problems).
Lipinski's Rule of Five states that, in general, an orally active drug has no more than one violation of the following criteria: • • • •
Not more than 5 hydrogen bond donors (nitrogen or oxygen atoms with one or more hydrogen atoms) Not more than 10 hydrogen bond acceptors (nitrogen or oxygen atoms) A molecular weight under 500 g/mol A partition coefficient log P less than 5
Note that all numbers are multiples of five, which is the origin of the rule's name.

Limitations of the rule-of-5 The rule-of-5 properties are not independent. Increasing MW often gives better potency due to more unspecific binding. However, increasing a compound’s size can by realized by adding more C or halogen atoms, leading to higher CLOGP, or by adding more hetero atoms, leading to higher hydrogen bonding capacity. Higher CLOGP, or lipophilicity, may result in poorer solubility. Higher H-bonding capacity may result in less membrane permeability. Optimizing on log D alone maybe insufficient as well. Since log D is a combination of size and hydrogen bonding, combinations of high MW and extensive H-bonding capacity are possible to yield a log D and solubility in the desired range. However, oral absorption of such compounds will nevertheless be limited because of the high H-bonding capacity. The balance between these properties begins to define the physicochemical space for success.

Brain targeting MW should be below 450 (most CNS drugs are relatively small in size, MW around 350), while for GI absorption a limit of 500 has been suggested. The total polar surface area, a measure for hydrogen bonding capacity, should be below 90. A well-known rule-of thumb that optimal CNS drug have a log D/P of ca.2!!! Octanol/water partition coefficients do not always correlate well with brain uptake solvent systems; alkane/water systems have been suggested (cyclohexane) it was demonstrated that the difference between octanol/water and alkane/water partition coefficients, Δlog P, gives satisfactory correlations with BBB crossing (alkane/water partition coefficient measurements are not simple, and often solubility limited).

Brain targeting: logP / logD In the fields of organic and medicinal chemistry, a partition (P) or distribution coefficient (D) is the ratio of concentrations of a compound in the two phases of a mixture of two immiscible solvents at equilibrium. Hence these coefficients are a measure of differential solubility of the compound between these two solvents (normally one of the solvents chosen is water while the second is hydrophobic such as octanol).

Brain targeting: PSA The Polar Surface Area (PSA) is defined as the surface sum over all polar atoms, (usually oxygen and nitrogen), including also attached hydrogen atoms. PSA is a commonly used medicinal chemistry metric for the optimization of cell permeability. Molecules with a polar surface area of greater than 140 angstroms squared are usually believed to be poor at permeating cell membranes. For molecules to penetrate the blood-brain barrier (and thus acting on receptors in the central nervous system), PSA should be less than 60 angstroms squared.

Farmakofór modellezés példa: antimaláriás hatású δ-kinolidin vegyületek vizsgálata Introduction Malaria is still one of the most dangerous infectious disease on the world. Despite the efforts toward to develop new agents, only a very few effective synthetic and natural antimalarial drugs were discovered in the past years, while the spread of resistance makes the old and cheap antimalarial agents ineffective. The main alkaloid of Cryptolepis sanguinolenta is cryptolepine (which constitutes about 1% of dried roots). Other quindoline structure based alkaloids were also isolated from this plant, they also shoved strong strong antiplasmodial activity agains P. Falciparum chloroquine-resistant strains, while quindoline was somehow less active. Cryptolepine itself failed to cure malaria in mice when given orally and was toxic when given by the intraperitoneal (i.p.) route. Cytotoxic properties are on account of its DNA-intercalating and topoisomerase II inhibiting activities as well as DNA synthesis. Fortunately, it appears that its antiplasmodial action (which like chloroquine involves the inhibition of haemozoin formation), is different (at least partially) from its cytotoxic mode of action, and therefore it may be possible to prepare analogues of cryptolepine that retain or have enhanced antiplasmodial activity but with reduced toxicity. Quantitative structure-activity relationship Quantitative structure-activity relationship (QSAR) (sometimes QSPR: quantitative structure-property relationship) is the process by which chemical structure is quantitatively correlated with a well defined process, such as biological activity (chemical reactivity) 3D-QSAR refers to the application of force field calculations requiring threedimensional structures, e.g. based on protein crystallography or molecule superimposition. It uses computed potentials, e.g. the Lennard-Jones potential, rather than experimental constants and is concerned with the overall molecule rather than a single substituent. It examines the steric fields (shape of the molecule) and the electrostatic fields based on the applied energy function.

1D – 2D QSAR The 1D models consider only the gross-formula of a molecule (for example, alanine—C3H7NO2). Actually, such models reflect only the composition of the molecule. Obviously, it is impossible to solve adequately ‘‘structure-activity’’ tasks using such approaches. These models have an auxiliary role. The 2D models contain information about the structure of the compound and are based on its structural formula. Such models reflect only the topology of the molecule. These models are very popular. The capacity of such approaches is that the topology model of molecular structure in an implicit kind contains information about possible conformations of the compound.

3D – 4D QSAR The 3D QSAR models give the full structural information, taking into account composition, topology and spatial shape of the molecule for one conformer only. These models are the most widespread. However, the choice of the analyzed conformer is mostly accidental. The most adequate description of molecular structure will be given by 4DQSAR models. These models are similar to 3D models, but as compared to them, the structural information is considered for a set of conformers (conditionally, the fourth dimension), instead of one fixed conformation.

Judging the quality of QSAR models The success of any QSAR model depends on: • accuracy of the input data • selection of appropriate descriptors and statistical tools • validation of the developed model Different aspects of validation of QSAR models that need attention includes methods of selection of training set compounds, setting training set size and impact of variable selection for training set models for determining the quality of prediction. Development of novel validation parameters for judging quality of QSAR models is also important.
