Peranan Retinoid Acid Reseptor Β2 (RAR Β2) Pada Proses Terjadinya Kanker Payudara Melalui Pendekatan Epigenetik Muliartha1), Wibi Riawan1), Fitri Armenia1), Asnah1) 1) Lab Biomedik Fakultas Kedokteran Universitas Brawijaya Malang Abstrak Penelitian ini bertujuan untuk mengungkap peranan gen retinoid acid reseptor (RAR β2) pada proses terjadinya kanker payudara. Peranan gen reseptor β2 retinoid adalah sebagai gen tumor supresor. Retinoid secara struktur dan fungsionil adalah vitamin A. Hilangnya ekspresi gen β2 asam retinoid reseptor (RAR β2) pada kromosom 3P24 salah satu faktor penyebab timbulnya kanker payudara. Sampai saat ini penyebab kematian yang nomor dua tertinggi pada wanita adalah kanker payudara. Pendekatan yang sekarang dipakai yaitu pendekatan genetika molekuler (Gen Apoptosis, Oncogen, Gen Supresor, Gen Growth Promoting Factor). Akan tetapi pendekatan tersebut belum bisa menuntaskan masalah. Penelitian yang akan dilakukan ini untuk mengungkap masalah tersebut. Kami merancang penelitian dengan desain eksperimental murni invitro dengan cara observasional. Sampel penderita kanker payudara berupa jaringan operasi tumor payudara yang sudah didiagnosa Infiltracting ductal carcinoma mama secara histopathology oleh seorang pathologis dengan teknik hematocylin eosin. Sebagian kecil hasil operasi tumor payudara ditumbuhkan dalam proses kultur jaringan. Identifikasi ekspresi gen reseptor retinoid acid β2 (RAR β2) dilakukan pada berbagai grading dari sel kanker payudara dengan teknik immunohistokimia. Untuk menganalisa hilangnya ekspresi RAR β2 pada kromosom 3P24 dilakukan dengan teknik PCR dilanjutkan dengan imunoblooting. Hasilnya terjadi penurunan ekspresi RAR β2 pada peningkatan grading dari kanker payudara. Terbukti dengan terjadi hilangnya RAR β2 pada kromosom 3P24 pada kanker payudara. Kesimpulan : pada kanker payudara memang terjadi proses penurunan ekspresi RAR β2 pada peningkatan grade serta menghilangnya RAR β2 pada kromosom 3P24 pada kanker payudara. Kata kunci : RAR β2, kanker payudara, patomekanisme.

Abstract The aim of this researche was to prove that gen retinoid acid reseptor (RAR β2) on the chromosom 3P24 to role of the pathogenesis breast cancer. The breast cancer is the most tumor malignant the second cause of death among the female. The recent concept of the theory of pathogenesis breast cancer, is the genetic moleculer approach. For example Apoptosis Gen, Oncogen, Supresor Gen, Growth Promoting Factor Gen, but the diagnostic and therapy of the breast cancer still not satisfaction. The lost or decrease expression of the gen RAR β2 on 3P24 chromosom are role of pathogenesis of the breast cancer. Design of the research true experimental observational invitro. Human female breast cancer sample from the deputy of surgery. Result : identification expression retinoid acid reseptor (RAR β2) shown by increase grade of the breast cancer and identified by immunohisto chemistry technic. Analyze the lost surpressor retinoid acid reseptor β2 (RAR β2) on the breast cancer identified by PCR continue by immunoblooting. Conclusion : identification expression of the RAR β2 shown by the increase grading of the breast cancer also show lost of the RAR β2 on 3P24 chromosom on the breast cancer.
