Enzymes zijn specifieke katalysatoren (8.1) De peptide binding is thermodynamisch labiel maar kinetisch zeer stabiel (half-waardetijd 1000 jaar).
De reactie die gekatalyseerd wordt door een proteolytisch enzym
De specificiteit van enzymen Trypsine en thrombine zijn beide proteases. Trypsine • Trypsine breekt eiwitten in de darmen af. • Thrombine werkt alleen in op één eiwit: fibrinogeen. Thrombine zet fibrinogeen in het bloed om in fibrine dat vervolgens polymeriseert en een stolsel vormt.

Koolstofdioxide wordt in de rode bloedcel gehydrateerd. Het gevormde bicarbonaat wordt uitgewisseld voor chloride en zo via het bloed naar de longen vervoerd. Het CO2 en de verlaging van de pH stimuleren de afgifte van zuurstof van hemoglobine tot 90% van zijn maximale transportcapaciteit.
Koolzuuranhydrase is een van de snelste enzymen. Elk enzymmolecuul kan per seconde 106 CO2 moleculen per seconde hydrateren. De katalytische cyclus wordt in een-miljoenste seconde doorlopen.
Veel enzymen hebben een cofactor nodig voor hun activiteit Apoenzyme + cofactor = holoenzyme Cofactoren die uit (kleine) organische moleculen bestaan noemt men coenzymen. Als het coenzymen stevig gebonden is noemt men deze een prosthetische groep. Wanneer het coenzym alleen tijdens de katalyse gebonden is noemt men deze een cosubstraat. Metalen zoals Zn en Ni zijn altijd stevig gebonden.

Enzymen kunnen energie in een andere vorm omzetten • Fotosynthese: licht(energie) in een chemische binding (NADPH en ATP). Uiteindelijk in gereduceerde koolstofverbindingen (glucose, vetzuren). • In mitochondriën wordt energie aanwezig in voedsel via redoxreacties omgezet in een ionengradiënt die wordt gebruikt om ATP uit ADP en fosfaat te vormen. • De bruikbare vrije energie die in ATP aanwezig is, wordt door enzymen gebruikt voor de contactie van spieren, voor actief transport van moleculen of voor de synthese van moleculen.

Wanneer Δ G0' = -5.71 kJ.mol-1, dan verschuift het evenwicht met een factor 10 naar rechts!!! Waterstofbindingen hebben een energie-inhoud varierend van 4 tot 20 kJ.mol-1
De glycolyse (figuur 16-2)
De omzetting van dihydroxyaceton fosfaat naar glyceraldehyde 3-fosfaat wordt nader thermodynamisch bekeken.
Bereken het vrije energie verschil voor de isomerisatie van DHAP naar GAP In evenwicht is de verhouding GAP/ DHAP 0.0475 bij 25 °C en pH 7. De standaard vrije energie wordt dan: • ΔG0' = -2.303 x R x T x log K'eq = -2.303 x 8.315 x 10-3 x 298 x log (0.0475) = 7.55 kJ mol-1 • Conclusie: de reactie is onder standaard condities een endotherme reactie.
• Wat gebeurt er wanneer de cellulaire concentraties worden gebruikt in de berekening? • DHAP = 2 x 10-4 M en GAP = 3 x 10-6 M? • ΔG' = 7.55 + 2.303 x RTlog([GAP]/[DHAP]) = 7.55 + 5.71 log(3x10-6/2x10-4) = 7.55 + 5.71 log (1.5x10-2) = 7.55 – 10.42 = -2.87 kJ mol-1 • The reaction is exotherm.

Het vrije energie verschil tussen de producten en dat van de substraten bepaalt de richting van de reactie (ΔG). Het vrije energieniveau van de overgangstoestand bepaalt de snelheid van de reactie (ΔG‡).

The significance of kcat - kcat of an enzyme is the number of substrate molecules that is converted per second into product per enzyme molecule under saturating substrate concentrations - kcat is also called the turnover number. Outline of chapter 15 • Metabolism is composed of many coupled and interconnected reactions • ATP is the universal currency of free energy in biological systems • The oxidation of carbon fuels is an important source of cellular energy (redox reactions) • Metabolic pathways contain many recurring motifs (the unifying themes of biochemistry)
Living organisms require a continual input of free energy for: • the performance of mechanical work in muscles and other cellular movements • active transport of molecules and ions • the synthesis of macromolecules

Structuur van het ATP synthase enzym complex • Vast deel (a, b2, δ, α en β subunits) • Draaiend deel (c ring en γ en ε subunits) • De γ subunit verbreekt de symmetrie van het α3β3 complex. • De α en β subunits binden beide nucleotiden, maar alleen de β subunit is betrokken bij de katalyse.
Hoe werkt het ATP synthase enzym?
Toevoegen van ADP en fosfaat geeft ATP dat zeer stevig aan het ATP synthase is gebonden. Door deze stevige binding wordt de reactie richting ATP synthese omgetrokken. Hoe drijft een proton gradiënt de ATP synthese, het loslaten van zeer stevig gebonden ATP?
Proton transport door het ATP synthase complex zorgt voor het loslaten van stevig gebonden ATP via een conformatie verandering van de β subunit.

Het proton pad door de membraan. • De hydrofobe a subunit bevat hydrofiele kanalen die niet met elkaar in verbinding staan. • De hydrofobe c subunit bevat in het midden een aspartaat residu. • Het aspartaatresidu kan geprotoneerd worden via een hydrofiel kanaal (protonrijke kant) van een a subunit. • Door te draaien kan het proton van de c subunit worden afgegeven aan het andere hydrofiele kanaal in een a subunit (proton-arme kant). • De c subunits draaien en daarmee via de γ subunit de αen β-subunits (ATP synthese).
Stoichiometry • Er zijn tussen de 10 en 14 c subunits per ATP synthase molecuul. • Als de γ subunit 360 graden gedraaid heeft dan zijn er 3 ATP moleculen gesynthetiseerd. • 10 c subunits per rondje 10 H+ ionen transporteren. H+ /ATP = 10/3 = 3.3. Per ATP molecuul worden 3 protonen verplaatst.
