Lampiran 10. Prosedur Standar Potong Beku
1. .Taringan dibekulcan dengan nitrogen cair. 2. Pastikan Cryostat bekerja pada temperatur -20°C sampai 30°C. Selanjutnya,
tempatkan sedikit OCT pada wadah yang telah berisi jaringan. (Gelembung tidak boleh terdapat pada sediaan) 3. Tempatkan spesimen yang telah dibekukan kebagian tengah cryobar pada cryostat
untuk proses pembekuan. Blok spesimen akan berwama putih solid saat telah membeku. 4. Bila spesilnen telah beku sempuma, tempatkan sedikit cairan OCT pada tengah disk
object untuk merekatkan blok spesimen. Selanjutnya tempatkan kembali pada cryobar, hingga spesimen menempel sempuma.
5. Apabila telah sempuma, tempatkan object disk yang telah ditempeli spesimen pada object disk holder dan dieratkan dengan memutar skmp objec dik holder.
6. Pastikan ada cukup jarak antara blok spesimen dengan pisau nzicrolome dan atur tebal spesimen yang akan dipotong dengan memutar pengatur tebal spesimen pada cryotome.
7. Gerakkan blok spesimen kearah pisau microtolne dengan memutar penggerak blok
pada bagian luar cryostat. Potong dan buang beberapa potongan pertama.
8. Selanjutnya ambil beberapa spesimen yang dibutuhkan dan rekatkan pada gelas objek. 9. Proses selanjutnya adalah melakukan pewamaan dengan Sudan Black dan Kemechtrot.
