[email protected] Fakultas Peternakan, Universitas Brawijaya, Malang Abstract Abstract Problems often arise in the process of freezing semen is sperm plasma membrane damage, because the membrane contains unsaturated fatty acids are highly susceptible to peroxidation damage. Formation Rective Oxygen Species (ROS) due to damage to the membrane causes a decrease in sperm quality. The purpose of this study was to analyze the supplementation of α-tocoferol in the diluent to maintain the quality of frozen semen to be used for Artificial Insemination (AI). This study used fresh semen Etawah Cross Breed Ram 2 to 2.5 years of age are accommodated once per week with an artificial vagina. Treatment research is the addition of antioxidants α-tocoferol into 100 ml of diluent with a dose of 0, 0.2, 0.4 and 0.6 g. Data were analyzed with randomized block design followed by Duncan's Multiple Range Test. The variables measured were quality of spermatozoa during cooling and thawing again, membrane integrity, the status of capacitation and acrosome reaction, percentage of intact plasma membrane and lipid peroxidation (malonaldehyde or MDA).The results showed that the addition of α-tocoferol 0.4 g/100 ml diluent to give the best results at all stages of freezing (p <0.05), so obtained83,33% pregnancy rate after synchronization of estrus with a double dose of prostaglandin (PGF2α) and IB first. The conclusion of this study is the dose α-tocoferol 0.4 g/100 ml diluent to maintain optimum quality of frozen Etawah Cross Breed Ram. AI applications on Goat PE females were synchronized to produce 76% pregnancy rate. Abstrak Masalah yang sering timbul dalam proses pembekuan semen adalah rusaknya membrane plasma spermatozoa. Pembentukan rective oxygen species (ROS) akibat rusaknya membrane spermatozoa menyebabkan penurunan kualitas spermatozoa. Tujuan penelitian ini adalah untuk menganalisis suplementasi α-tocoferol dalam pengencer terhadap kualitas semen beku yang akan digunakan untuk inseminasi buatan (IB). Penelitian ini menggunakan semen segar kambing Peranakan Etawah (PE) umur 2-2,5 tahun. Perlakuan penelitian adalah penambahan α-tocoferol kedalam 100 ml pengencer AndromedRdengan dosis 0,0; 0,2; 0,4 dan 0,6 g. Data dianalisis dengan analisis varian dan dilanjutkan dengan uji Jjarak berganda Duncan. Variabel yang diamati adalah kualitas spermatozoa pada saat pendingin dan pencairan kembali, yang meliputi integritas membrane, status kapasitasi dan reaksi akrosom, persentase membrane plasma utuh serta kadar lipid peroksidase (Malonaldehid atau MDA). Hasil pengamatan menunjukkan bahwa penambahan α-tocoferol 0,4 g/100 ml pengencer memberikan hasil terbaik pada semua tahapan pembekuan (p<0,05), sehingga didapatkan angka kebuntingan 83,33% setelah sinkronisasi birahi dengan dobel dosis prostaglandin (PGF2α) dan IB pertama.
