Lampiran 1. Prosedur Analisa 1. Vitamin C (Sudarmadji et al, 1997) Kandungan Vitamin C dihitung dengan metode titrasi / Jacobs. Sebanyak 10 g sampel dilarutkan dengan aquades dalam labu takar 100 ml kemudian disaring. Setelah itu, sebanyak 10 ml larutan sampel ditetesi dengan indikator pati sebanyak 2-3 tetes dan dititrasi dengan menggunakan larutan iod 0,01 N. Titik akhir titrasi ditandai dengan perubahan warna larutan biru. Tiap ml iod equivalen dengan 0,88 mg asam askorbat. Kadar vitamin C dalam contoh dihitung dengan rumus : C=
Keterangan : C = mg asam askorbat / 100 gram contoh FP = faktor pengenceran 2. Persen (%) Total Asam pada buah-buahan (Rangana, 1979). Penentuan total asam dengan cara titrasi. Sebanyak 10 ml sari buah dipipet ke dalam labu erlenmeyer 100 ml dan ditambahkan air destilasi sebanyak 15 ml serta ditetesi indikator phenolphtalein. Kemudian dititrasi dengan larutan NaOH 0,1 N sampai diperoleh warna merah jambu. Total asam dihitung dengan menggunakan rumus : TA = Dimana : TA = total asam (mek/100 ml sari buah) V = volume larutan NaOH (ml) N = Normalitas larutan NaOH (ml) P = jumlah pengenceran

3. Kejernihan Kejernihan diukur dengan menggunakan spektrofotometer, sebanyak 0,1 ml sari buah dipipet kedalam tabung reaksi dengan menggunakan mikropipet kemudian 0,9 ml aquades ditambahkan.
Sampel dikocok dan dimasukkan
kedalam spektofotometer yang menggunakan panjang gelombang 420. 4. Bilangan formol (SNI 01-2891-1992) Pengujian bilangan formol dilakukan berdasarkan pada, cara uji makanan dan minuman butir 4,4. Sebanyak 10 ml sari buah ditambahkan dengan 10 ml aquadest, kemudian dinetralkan dalam gelas piala dengan NaOH 0,25 N sampai pH 8,1 dengan menggunakan pH meter.
Ditambahkan 10 ml larutan
formaldehida. Setelah kurang dari 1 menit larutan dititar dengan NaOH 0,25 N sampai pH 8,1.

Keterangan : a= ml NaOH 0,25 N yang dipergunakan untuk titrasi b= Normalitas NaOH yang dipergunakan untuk titrasi c= volume contoh 5. Viskositas (cp) Viskositas (cp) sari buah rambutan diukur dengan menggunakan viskometer Brookfield pada tingkat kecepatan pengadukan dan spindle yang disesuaikan dengan sampel. Nilai viscositas yang terbaca dinyatakan dalam centipoise. 6. Total Padatan Terlarut (oBrix) Total padatan terlarut diukur dengan alat Refraktometer. Sampel sari buah diteteskan pada refraktometer dan nilainya dibaca pada skala oBrix yang tertera. Refraltometer yang digunakan menggunakan selang 0-30 %.

7. pH (Apriyantono, 1989) Elektroda dari pH meter dicelupkan ke dalam larutan buffer (penyangga) terlebuh dahulu untuk kalibrasi alat. Kemudian dicelupkan ke dalam larutan sampel yang akan dianalisis keasamannya (pH) berapa nilai pH-nya akan tertera langsung pada layar digital pH meter tersebut. 8. Total Mikroba Sari buah/Total Plate Count (Fardiaz, 1989) Sebanyak 1 ml sampel dimasukkan ke dalam 9 ml larutan pengencer. Selanjutnya dilakukan pengocokan hingga homogen dengan vortex. Pengenceran dan pemupukan dilakukan dengan tingkat pengenceran 10 -2. Dari tiap-tisp pengenceran, dipipet secara aseptic 1 ml untuk dimasukkan ke dalam cawan petri steril (pemupukan) secara duplo dan ditambahkan media NA (Nutrient agar) steril sebanyak 15-20 ml. Segera setelah penuangan, cawan petri digerakkan di atas meja secara hatihati untuk meyebarkan sel mikroba secara merata, yaitu dengan gerakan melingkar atau angka delapan.
Setelah media membeku, cawan petri
diinkubasikan dengan posisi terbalik pada incubator suhu 37oC selama 1 hari (12 jam).
Perhitungan jumlah total mikroba dilakukan dengan menggunakan
Standard Plate Count (SPC) metode Harrigan. 9. Uji organoleptik (Soekarto, 1985), Uji organoleptik dilakukan dengan metode consumer preference test atau uji kesukaan konsumen yaitu menggunakan panelis terlatih sebanyak 15 orang. Bahan disajikan secara acak dengan diberi nomor kode, kemudian panelis diminta untuk memberikan penilaian pada salah satu kriteria skala hedonik. pengamatan dinyatakan dengan 7 skala hedonik, berikut : 1. 2. 3. 4. 5.
