Lampiran 1. Prosedur analisis proksimat minyak biji kamandrah Rendemen Minyak Rendemen dihitung dengan membandingkan jumlah minyak yang dihasilkan dari bahan baku yang diproses.

Kadar Air Cara Pemanasan (AOAC, 1996) Cawan kosong dan tutupnya dikeringkan dalam oven selama 15 menit dan dinginkan dalam eksikator, kemudian ditimbang (untuk cawan alumunium didinginkan selama 10 menit dan cawan porselen didinginkan selama 20 menit). Timbang dengan cepat kurang lebih 5 gram sampel yang sudah dihomogenkan dalam cawan. Angkat tutup cawan dan tempatkan cawan berserta isi dan tutupnya di dalam oven selama 6 jam. Hindarkan kontak antara cawan dengan dinding oven. Untuk produk yang tidak mengalami dekomposisi dengan pengeringan yang lama dapat dikeringkan selama 1 malam (16 jam). Pindahkan cawan ke desikator, tutup dengan penutup cawan, lalu didinginkan. Setelah dingin timbang kembali. Keringkan kembali ke dalam oven sampai diperoleh berat yang tetap.
Perhitungan : Berat sampel (gram) = W1 Berat sampel setelah dikeringkan (gram) = W2 Kehilangan berat (gram) = W3 W3 Persen kadar air (wet basis) = x 100 W1 W3 Persen kadar air (dry basis) = x 100 W2
Kadar Lemak dan Minyak dengan Soxhlet (Woodman, 1971 dalam Sudarmadji dkk. 1997) Ambil labu lemak yang ukurannya sesuai dengan alat ekstraksi soxhlet yang akan digunakan, keringkan dalam oven, dinginkan dalam desikator dan
timbang. Timbang 5 gram sampel dalam bentuk tepung langsung dalam saringan timbel, yang sesuai ukurannya, kemudian tutup dengan kapas-wool yang bebas lemak. Sebagai alternatif sampel dapat dibungkus dengan kertas saring. Letakkan timbel atau kertas saring yang berisi sampel tersebut dalam alat ekstraksi soxhlet, kemudian pasang alat kondenser diatasnya, dan labu lemak dibawahnya. Tuangkan pelarut dietil eter atau petrolium eter ke dalam labu lemak secukupnya, sesuai dengan ukuran soxhlet yang digunakan. Lakukan refluks selama minimum 5 jam sampai pelarut yang turun kembali labu lemak berwarna jernih. Distilasi pelarut yang ada didalam labu lemak, tampung pelarutnya. Selanjutnya labu lemak yang berisi lemak hasil ekstraksi dipanaskan dalam oven pada suhu 105oC. Setelah dikeringkan sampai berat tetap dan didinginkan dalam desikator, timbang labu berserta lemaknya tersebut. Berat lemak dapat ditimbang.

Kadar Abu Cawan yang telah dibersihkan dipanaskan dalam tanur pada suhu 100oC selama 2 jam lalu ditimbang sebagai bobot kosong. Contoh yang telah diuapkan ditimbang teliti + 1g dalam cawan dan dinyatakan sebagai bobot awal, kemudian cawan tersebut dimasukkan ke dalam tanur suhu 600oC selama 5 jam. Setelah pemanasan cawan dimasukkan ke dalam desikator, dan setelah dingin ditimbang dan dipanaskan beberapa kali sampai diperoleh bobot tetap sebagai bobot akhir.

Kadar Protein
Sampel ditimbang secara teliti sebanyak 200 mg, lalu dimasukkan ke alam labu Kjeldhal. Selanjutnya ditambahkan selen dan 10 ml asam sulfat pekat dan didestruksi pada pemanas selama 2-3 jam atau sampai larutan menjadi jernih. Setelah proses destruksi lalu dipindahkan ke dalam labu destilasi kemudian diperiksa kandungan nitrogennya dengan menggunakan alat kjeldhal.
Perhitungan :
Keterangan: a= bobot contoh b= volume HCl yang digunakan 6,25 = faktor konversi dari nitrogen ke protein 14 = BM nitrogen
Serat Kasar Sampel yang telah digunakan pada penetapan lemak ditimbang dengan teliti + 500 mg lalu dimasukkan ke dalam erlenmeyer. Selanjutnya ditambahkan 100 ml asam sulfat 1,25% dan dipanaskan sampai mendidih. Setelah 1 jam ditambahkan 100 ml natrium hidroksida 3,25%, dipanaskan kembali sampai mendidih selama 1 jam, kemudian didinginkan dan disaring dengan menggunakan kertas saring yang telah diketahui bobotnya. Endapan dicuci dengan asam sulfat encer dan alkohol, lalu kertas saring dan endapan dikeringkan dalam oven dan ditimbang.

Lampiran 2. Prosedur analisis sifat fisiko-kimia minyak biji kamandarah Indeks bias Indeks bias, berdasarkan SNI 06-3735-1998, prinsip yang dipakai adalah jika cahaya datang dan menembus dua media dengan kerapatan yang berbeda maka akan dibelokkan atau dibiaskan mendekat atau menjauh garis normal. Cara Kerja : Prisma pada refraktometer dibersihkan dengan alkohol, kemudian diatas prisma ditetesan sampel menggunakan pipet tetes. Prisma dirapatkan dan diatur slidnya sehingga diperoleh garis batas yang jelas antara terang dan gelap, saklar diatur sampai garis batas berhimpit dengan titik potong dari dua garis bersilangan, indeks bias dibaca.
Perhitungan : Indeks bias (25oC) = n’ – 0,00045 (t – 25) Keterangan : t : suhu kamar (oC) n’ : indeks bias pada suhu kamar 0,00045 : faktor koreksi yang nilainya dapat diberubah sesuai dengan suhu yang dipakai.
Bobot jenis Bobot jenis adalah perbandingan berat dari volume sampel minyak dengan berat air yang volumenya sama pada suhu tertentu (biasanya ditentukan pada suhu 25oC).
Cara Kerja : Piknometer dibersihkan dan dikeringkan. Isi piknometer dengan aquades bersuhu 20-30oC. Pengisian dilakukan sampai air dalam botol meluap dan tidak
ada gelembung udara didalamnya. Setelah ditutup, botol direndam dalam bak air yang bersuhu 25oC dengan toleransi 0,2oC selama 30 menit. Botol diangkat dari bak dan dikeringkan dengan kertas pengisap. Timbang berat botol dengan isinya. Contoh minyak/lemak cair yang akan ditentukan berat jenisnya, sebelum disaring dulu dengan kertas saring. Hal ini bertujuan untuk membuang benda-benda asing dan kandungan air. Selanjutnya contoh minyak diperlakukan seperti langkah 1 sampai dengan 5.
Perhitungan : Berat botol dan minyak – berat botol Berat jenis minyak pada suhu 25/25oC = Berat air pada suhu 25oC Jika berat minyak pada suhu 25oC telah diketahui maka untuk menghitung berat jenis minyak pada suhu tertentu lainnya dapat digunakan rumus :

Bilangan Asam (AOAC, 1995) Bilangan asam dinyatakan sebagai jumlah miligram KOH yang dibutuhkan untuk menetralkan asam lemak bebas yang terdapat dalam 1 gram minyak atau lemak. Bilangan asam menyatakan jumlah asam lemak bebas yang terkandung dalam minyak/lemak, biasanya dihubungkan dengan proses hidrolisa minyak/lemak yang berkaitan dengan mutu minyak/lemak.
Cara Kerja : Timbang 10-20 gram minyak/lemak dalam erlenmeyer 250 ml. Tambahkan 50 ml alkohol 95% netral, panaskan sampai mendidih (+ 10 menit) dalam penangas air sambil diaduk. Larutan ini kemudian dititrasi dengan KOH 0,1 N, menggunakan indikator fenolftalin sampai terbentuk warna merah jambu yang persisten selama 10 detik.
Perhitungan : ml KOH x N KOH x 56,1 Bilangan asam = Berat sampel (g)
ml KOH x N KOH x M Kadar asam = 10 x Berat sampel (g) Keterangan : M = berat molekul asam lemak yang dominan dalam minyak/lemak (Contoh untuk oleat = 282)
Kadar Asam Lemak Bebas (Persen FFA) Bilangan asam sering juga dinyatakan sebagai kadar asam lemak bebas (% FFA). Hubungan kadar asam lemak bebas dengan bilangan asam menurut Sudarmadji dkk (1989) dapat dituliskan sebagai berikut : Perhitungan : Bilangan asam % FFA = Faktor konversi
Keterangan : Faktor konversi untuk oleat = 1,99 Faktor konversi untuk palmitat = 2,19 Faktor konversi untuk laurat = 2,80 Faktor konversi untuk linoleat = 2,01
Bilangan Peroksida (AOAC, 1995) Penentuan bilangan peroksida biasanya didasarkan pada pengukuran sejumlah iod yang dibebaskan dari potasium iodida melalui reaksi oksidasi oleh peroksida dalam lemak/minyak pada suhu ruang didalam medium asam asetat/kloroform.
Cara Kerja : Ditimbang 5 gram contoh minyak ke dalam erlenmeyer 250 ml. Tambahkan 30 ml pelarut (60% asam asetat glasial dan 40% kloroform), kocok sampai semua contoh minyak larut. Tambahkan 0,5 ml larutan potasium iodida (KI) jenuh,
diamkan selama 2 menit diruang gelap sambil digoyang. Tambahkan 30 ml air destilate. Tambahkan indikator pati dan kelebihan iod dititrasi dengan larutan sodium tiosulfat (Na2S2O3) 0,1 N atau 0,01 N tergantung dari banyaknya iod yang dibebaskna. Dengan cara yang sama buatlah penetapan untuk blanko

Lampiran 3. Prosedur uji Larvasida Penyiapan larva nyamuk Ae. aegypti Telur nyamuk Ae. Aegypti dicelupkan dalam air suling pada nampan plastik ukuran 34 x 25 x 4,5 cm sebanyak 1 liter, telur akan menetas setelah + 24 jam menjadi larva instar I. Larva dipelihara dengan pemberian pakan hati ayam yang telah dikukus dan dihaluskan hingga menjadi instar III selama 3-4 hari. Perubahan tiap instar ditunjukkan dengan terjadinya ecdysis (pelepasan kulit).
Penyiapan sampel Sampel dibuat seri konsentrasi dari 0 ppm, 10 ppm, 50 ppm, 100 ppm, 250 ppm, dan 500 ppm, kemudian dihomogenkan.
Pelaksanaan Uji Sampel dari setiap seri konsentrasi diambil 200 ml dan dimasukkan dalam gelas plastik, kemudian masukkan 25 ekor larva Ae. Aegypti instar III. Ulangi perlakuan ini untuk 5 ulangan.
Pengamatan Pengamatan dilakukan setelah 24 dan 48 jam dengan menghitung berapa larva yang mati.
Analisis Data Untuk menentukan angka kematian 50% dan 90% (LC50 dan LC90) dapat dilakukan dengan metode probit analisis (Finney Method) dengan menggunakan
software POLO-PC (Le Ora Software 1987) atau software SPSS versi 17.
