Perlindungan d m Efek Penyembuhan Sediaan Bangle (Zingiber cmmmwwr Roxb) terhadap Peradangan Hati Tikus serta Mekanismenya pada Sel Makrofag dan Lirnfosit Di bim bing oleh DEDDY MUCHTADI, FRANSISKA RUNGKAT ZAKARIA, TUTIK WRESDIYATI dm SIDIK. Rimpang bangle (Zingiber cassumunar Roxb) telah digunakan sebagai obat tradisional dan rempah seperti halnya hnyig dan telah dikctahui mempunyai aktivitas antioksidan dan antiperadangan. Penelitian ini bertujuan menguji kemampuan sediaan bangle dalam melindungi dan menyembuhkan hati tilcus terhadap peradangan akibat induksi CC4 dan rncrnpelajari rnekanisme antiperadangan dim bangle secara in vim ~ pada n u h o edan limfosit. Penelitian ini dibagi dalam 3 tahap. Tahap pertama adalah karakterisasi sediaan uji SK, m i EA dm fraksi KLT. Karakteristik yang diamati adalah penampakan fisik, w m a dan amma, rendemen, kadar total fenol dm aktivitas penangkapan d i k a l bebas. Tahap kedua (in vivo). Pada tahap ini, 175 ekor tikus Sprague-Dawley jantan berumur 8 minggu dibagi rnenjdi 7 kelompok (n=25), yaitu kelompok yang mendapatkan fraksi etil asetat 15.00 m@g BB (EAl), 30.00 mfig BB (EA2), 60.00 mgkg BB (EA3); kelompk yang mendapat sari bangle kering 30.00 mglkg BB (SK);kelompok yang dan mendapt kurkuminoid 30.0 rng/kg BB (KUR); kelornpok kontrol negatif kelornpok kontml (K) mendapat 5% lamtan T w n M) (pelarut sediasn uji). Semua tikus mendapatkan pakan standar dm minum ad libitum. Semua tiicus, kecuali tikus kelompok KN dan K, diberi sediaan uji selama 7 hari, secara oral menggunakan sonde. Pada hari ke-7 semua tikus, kecuali tikus kelornpok K, dipaparkan dengan CC14 0.1 mVkg BB per oral. Kemudian, ke-7 kelompok masing-mslsing dibagi menjadi 5 sub keIornpok (n=5) berdasarkan Iamanya waktu pemaparan CC14yaitu 3 jam, 24 jam, 48 jam, 72 jam, dan 96 jam. Setelah pemMan CC4 kelompok EA1, EA2, EA3, SK, KUR tetap diberikan sediaan uji clan pakan standar, sedangkan kelompok KN dan R tetap diberikan pelarut sediaan uji dan perkan standar. Tahap ketiga penelitian addah pendugaan mekanisme antipedangan SK, h k s i EA dm fraksi KLT secara in vim. Penelitian ini mempelajari p e n g a d penarnbahan sediaan uji terhadap kadar NO dan PGE2 pztda kuItur sel m h f a g RAW 264.7, dan LTB4 pada sel limfosit tikus. Sediaan SK dan W i EA3 dapat melindungi hati tikus dari p e r h g a n &bat panaparm CC4, Hal ini didasarkan pada kemampuan sediaan SK dan fraksi EA3 berturu-tumt memkrikan kadar SGOT dm SGPT pada selang normal serta MDA yang mendeekati normal pada 3 jam setelah pernapman CC4, Fraksi EA3 mempunyai efek penyembuhan peradangan hati terbaik dibandingkan dengan sediaan SK, fraksi EA1 dan EA2. Selain itu, m i W juga mampl mmingbtlrao jumlah a 1 y m g positif PCNA, yang rnenunjukkan kemarnpuan menstimulasi proliferasi sel. Fraksi EA mempunyai efektifitas yang -a d e n p indomctscin dalam menckan Mar NO. Pada kadar 10.00 pg/ml fmksi EA rnempunyai k c m q u m menekan kadar PGEZ yang lcbih baik dibandigkem SK dan KLT. Pada kadar 0.01 pglml masing-masing sediaan SK, fraksi EA, dan h k s i KLT mempunyai kemampuan yang tidak berbeda nyata dengan indometach dalam maghambat kadar LTB4.

ELMEIZY ARAFAH. Protective and Curative Effects of Bangle (Zingiber cussumunar ROXB) Rhizome Extracts on Liver Inflammation in Rats and Their Mechanisms on Macrophage and Lymphocyte Cells. Under the direction of Deddy Muchtadi, Fransiska R Zakaria, Tutik Wresdiyati and Sidik. Zingiber cCISmmunar Roxb, known as bangle, has antioxidative and antiinflammatory activities and has been used for medicinal purposes traditionally. The aims of this research were to investigate whether administration of 2. cassumunar extracts orally may prevent acute liver damage induced by carbontehachloride (CC4) in rats and to elucidate the mechanisms of the anti-inflammation on macrophage and lymphmyte in v i m . Characterization of h e z e dried bangle juice (SK), ethyI acetate fraction P A ) , and TLC W o n , included physical properties, color and aroma, total phenolic compounds and radical scavenging activity of the extracts. Evaluation of protective and curative effect of freeze dried bangle juice and ethyl acetate fraction in CC4 induced Sprague-Dawley male rat liver. The 175 male Sprague-Dawley rats (8 months) were divided in 7 groups (n=25). Group 1,2, and 3 were given ethyl acetate sadion 15.00 mglkg bw (EAI), 30.00 mgntg bw (EA2), or 60.00 mgntg bw (EA3). Group 4 was given h e z e dried bangle juice 30.00 mglkg bw (SK), group 5 was given curcuminoid 30.00 mgkg bw (KUR), group 6 as negative control (KN) and group 7 as control (K) were given extract solvent. Each group, except KN and K groups were given the extracts for 7 days by oral using sonde. Carbontetrachloride 0,l mYkg bw was. given orally after 7 days of treatments to group EAl, EA2,EA3, SK, KUR and KN. After receiving CC14, all groups were continued given treatment until termination, After that, each group was divided to 5 sub groups (n=5). The subgroup were sacrificed 'after 3,24,48,72, and 96 hours after receiving CC4. The effects of extracts on the anti-inflammation mechanism was done wing in vim method. This mge was done by evduating the effect of SK,EA and TLC fraction e e t s on nitric oxide and prostaglandine & on murine macrophage R A W 264.7 and leucotriene B4on rat lymphocyte production The SK and EA3 k t i o n s could protect rat liver h m -inflammation by CCLinduced, as shown by their SGOT, SGPT a d MDA level which were near to noma1 Ievel. The EA3 W o n given to rats 60.00 mgkg bw had curative effect on rat liver inflammation through decreasing SGOT and SGPT level, MDA liver. This extract seemed to stimulate liver recovery by increasing .prolifersting PCNA positive cells. The SK, EA M o n and TLC fraction of bangle rhizomes could inhibited NO, PGEz and LTB4 level depending on their concentrations. The EA fraction had the same affdvities with i n d o m a i n for inhibiting NO Ieve1. The EA hction could reduced significantly PG& level compare to SK and TlLC fraction at concentration of 10.00 pglml. The SK, EA M i o n and TLC fraction had no significant difference with indometacin on LTB4 level at concentration 0.01 pg/ml.
